Outline of Annual Research Achievements

本研究の目的は,水溶液中の柔らかな分子系のダイナミクスを観測する超高速近赤外振動分光計を開発し,発色団とタンパク質との選択的結合によって発色団の動力学が受ける影響を明らかにすることである.本年度はアスタキサンチン結合タンパク質の誘導ラマンスペクトル測定を目指したが,タンパク質の単離がきわめて難しく,測定に至らなかった.そこで,アスタキサンチンおよびβ-カロテンの有機溶媒溶液について時間分解近赤外誘導ラマンスペクトルを測定し,カロテノイドの緩和ダイナミクスを詳細に検討した.
まず,アスタキサンチンのフェムト秒時間分解近赤外吸収および誘導ラマンスペクトルを測定した.近赤外吸収の強度の時間変化から,S2状態からS1状態への内部転換の時定数を0.15 psと見積もった.また,S1状態のC=C伸縮振動バンドの高波数シフトから,S1状態における振動エネルギー緩和の時定数を0.3 psと見積もった.
アスタキサンチンについて得られた内部転換および振動エネルギー緩和の時定数を,β-カロテンについて得られた時定数と比較した.アスタキサンチンの内部転換および振動エネルギー緩和の時定数はそれぞれβ-カロテンの0.8倍,0.33倍であった.末端基のわずかな違いにより,カロテノイドの緩和の時定数が大きく変化することが示された.
光励起の際にアスタキサンチンに余剰なエネルギーを与え,余剰エネルギーがアスタキサンチンの内部転換や振動エネルギー緩和に及ぼす影響を調べた.余剰エネルギーが増大するとS2状態のC=C伸縮振動のピーク波数が低くなった.ピーク波数の時間変化を調べた結果,光励起の際に与えた余剰エネルギーは,S2状態の寿命の時間範囲内には散逸しないことが分かった.また,この結果がカロテノイドの末端基の影響を受けないことが明らかとなった.
