Outline of Annual Research Achievements

ラベルフリーで細胞3Dセンシング操作を実現するために、ひとつの断面に16電極を5断面合計80電極を内層させた菱形のマイクロ流路を作製した。(1)細胞と流体との固液二相流状態で流路内部のインピーダンスをリアルタイムで計測することにより、細胞の電気的特性のセンシングを行った。具体的には、EIS法を用いて、異なる体積濃度を持つ細胞懸濁液の誘電特性を計測し、Maxwell-Wagner分散理論と細胞分極モデルを用いて、計測した誘電特性を解析した。さらに、マイクロ流路内における誘電特性を用いた細胞の空間的濃度分布計測について検討した。EIT法を用いてマイクロ流路内における細胞濃度分布の画像化を行なった。EIS法とEIT法との結果を比較し、細胞の空間的濃度分布計測について検討した。この結果により、EIT法を用いて非侵襲かつオンラインでの細胞濃度断面3D可視化計測が可能となった。(2)流路内部の電極に比較的周波数の高い電流を流し、誘電泳動力などによる細胞挙動のPIV計測を行った。具体的には、3種類の電気的特性が異なるMRC-5 Wild type、細胞核内に正電荷を有するGFP-histone 2Bを大量に発現させたMRC-5 GFP-histone 2B expressing type、細胞質内に負電荷を有するGFPが発現したMRC-5 GFP expressing typeを用意した。それらの3種類の細胞をD-PBSに懸濁し、それぞれの細胞懸濁液をマイクロ流路内に充填させ、マイクロ流路内電極へ交流電圧を印加したときの細胞挙動のPIV計測を行った。その結果、電極近傍で3種類細胞の移動速度関係が明らかとなり、細胞膜内部および細胞核内部に存在する電荷を有する物質量の違いを利用した細胞の分離技術の確立に対して可能性を見出した。
