Outline of Annual Research Achievements

本年度は、エクソソームの定量解析を進めるために、(1)エクソソームに内包されるmicroRNA等の核酸を定量解析することが可能なナノデバイスと(2)エクソソーム表層の膜タンパク質を定量解析することが可能なナノデバイス
を開発した。申請者は昨年度までに、ナノワイヤにより抽出分離したエクソソームに内包されるmicroRNA等の核酸解析と、エクソソーム表層の膜タンパク質の解析に成功した。本年度は、そのナノワイヤ構造体を進化させることで、(1)溶液中の全エクソソームの抽出分離&内包されるmicroRNA等の核酸定量解析と、(2)表層にある全膜タ
ンパク質の定量解析、が可能な新規ナノデバイスを連続成長ナノワイヤ構造体を用いて創製した。(1)の課題においては、前年度に作製したデバイスにおいて完全抽出分離したエクソソームからmicroRNA等の核酸が全て抽出されるかどうかを検討した。その結果、抽出されたmicroRNA量と抽出分離されたエクソソーム量が一致した(従来法では15%程度に対し、本手法では95%以上の抽出)。(2)の課題においては、ナノワイヤ上にエクソソームを捕捉した後に、CD9, CD63, CD81, CD147等のエクソソーム表層の膜たんぱく質に対して、蛍光標識化した抗体を用いて、抗原抗体反応にて検量線を作製し、定量解析を行うことに成功した。
