Outline of Annual Research Achievements |
生体分子の機能に影響を及ぼす因子の一つに、周辺の分子環境がある。細胞内は生体分子が非常に混み合った分子クラウディング状態にあるに対し、生化学的な実験が行われる試験管内環境は分子濃度が極めて希薄である。そのため、試験管内での実験結果をもとにして、細胞での生体分子の物性を解明することが困難であることが明らかになりつつある。 そこで本研究では、①細胞核の化学環境を模倣するために、領域内の共同研究者が設計した負電荷高分子を用いて核内に高濃度に存在するゲノムを模倣した。また、②細胞質を化学模倣するために、細胞質に数百mMから数M程度も存在する浸透圧調節分子を用いて細胞を化学模倣した実験系を構築した。これらの分子環境において、核酸の構造安定性を定量解析したところ、核酸の非標準構造が特異的に安定化されることが明らかとなった。③さらにこれらの知見をもとにして、四重らせん構造リガンドを細胞内で放出するシステムを構築した。 これらの成果の一部は、掲載論文誌の表紙として取り上げられ、新聞報道もされた。これらのことから、研究の最終年度として、科学的・社会的にインパクトのある成果が得られたといえる。また、本研究課題を発展させた成果も得られつつあることから、研究開始当初に設定した目標を十分に達成したといえる。
