Outline of Annual Research Achievements |
本研究では、これまでに解明されていない脳・神経系におけるコンドロイチン硫酸(CS)代謝分解機構を明らかにし、分解酵素による脳・神経系におけるCSの機能調節機構を調べようとしている。今年度は、(1)脳内におけるCSおよびヒアルロン酸(HA)分解酵素活性の検出、およびその酵素活性を担うタンパク質の同定、(2)CSおよびHA分解酵素の候補遺伝子の脳内における発現量の詳細な解析、(3)脳・神経に由来する培養細胞を用いた、CSおよびHA分解機構の解明、(4)脳・神経における主要なCSおよびHA分解酵素の性状・機能解析を目指した。(1)および(3)に関して、酵素活性の検出に成功した。それぞれからの酵素の精製を開始したが、(3)の培養細胞を用いる方が効果的に精製できると考え、(1)の脳組織からの酵素精製に関しては中断した。イオン交換クロマトグラフィーによって部分精製ができた。現在質量分析を用いたタンパク質の同定を行っている。(2)については、様々な酵素の発現解析ができ、ヒアルロニダーゼ2の高発現を観察した。ヒアルロニダーゼ2の貢献を確認するため、shRNAによるノックダウン細部株を樹立し、現在解析中である。(4)の項目については、(1)~(3)までで特定の分子の同定にまで至っていないため、解析を進めることはできなかった。また、上記以外に、CS特異的な加水分解酵素として我々が同定したHYAL4について、酵母でのHYAL4大量発現系の構築と酵素の大量調製、細胞内でのHYAL4の局在部位の解析、HYAL4の基質特異性に最も大きく貢献するアミノ酸残基の同定を行った。これらの研究成果の一部は、複数の学会で報告した。
