Outline of Annual Research Achievements

髄液に含まれる蛋白質と血清中の蛋白質では糖鎖修飾が大きく異なっており、糖鎖構造の多様性と神経疾患との密接な関わりがクローズアップされている。例えば神経疾患の一つである特発性正常圧水頭症(iNPH)はその簡便な診断法が確立されていなかったが、最近になり髄液中のトランスフェリンの糖鎖構造がiNPHの診断マーカーとなる可能性が報告された。申請者は平成24~25年度の公募研究から生まれた融合研究により、髄液中のトランスフェリンの部位特異的な糖鎖解析から、新奇な糖鎖構造であるN-GlcNAc型糖鎖を持つ画分が存在することを見出した(Nagae et al. Glycobiology 2014)。N-GlcNAc型糖鎖はマウスのシナプス後肥厚(Postsynaptic density, PSD)を対象にした最近のプロテオミクス解析でも見出されており、神経系に特有の糖鎖構造である可能性がある。しかしながら、N-GlcNAc型糖鎖の分布や、細胞外画分におけるN-GlcNAc型糖鎖の生成メカニズムは全くといっていいほど不明である。本申請課題は、髄液内に含まれる糖蛋白質の広範な解析によって、N-GlcNAc型糖蛋白質群を同定することを第一の目的とする。さらに第二の目的として髄液におけるN-GlcNAc型糖鎖の生成メカニズムを生化学的手法および情報科学的手法により解析をし、その生理的意義を明らかにする。我々は第一の目的に沿って、これまで質量分析を用いた髄液内の網羅的な糖蛋白質解析によりN-GlcNAc型糖蛋白質を新たに一種類見出した。また第二の目的について、N-GlcNAc型糖鎖の生成が細胞外領域で起きるという作業仮説の下、髄液中の遊離糖鎖の解析を行い、血清と比較して髄液中にはほとんどN型糖鎖の代謝産物は見られないことを見出した。また新規シアル酸代謝産物を同定した。
