Outline of Annual Research Achievements |
遺伝性膵炎の原因遺伝子SPINK1をマウスSpink3 locusに置換し、SPINK1ノックインマウスを樹立した (Spink3SPINK1/SPINK1)。SPINK1の変異により、①SPINK1の機能低下、②SPINK1発現量の低下、の2つの可能性がin vitroの実験から明らかにされている。そこで、②を検証するために、Spink3SPINK1/SPINK1とSpink3+/-マウスとの交配によりSPINK1ヘテロノックインマウス(Spink3SPINK1/-)を作製し、SPINK1の発現量が約70%低下していることを確認した。このマウスの致死性は回避されるものの、生後4週齢において、慢性膵炎(膵腺房細胞の脱落、線維化、炎症細胞浸潤)を発症した。 このマウスの膵炎発症過程において、オートファジーに由来する巨大な空胞の出現と、オートファジーの選択的基質であるp62タンパクの蓄積が見られた。つまり、SPINKの欠損、および発現低下はオートファジーの異常を引き起こすことで、膵炎を発している可能性が示唆された。さらにこのSpink3SPINK1/-マウスでは、膵腺房細胞がアポトーシスとネクローシスによって脱落していることが明らかとなったが、特にプログラムされたネクローシス(ネクロプトーシス)の実行因子であるRIP3を欠失させたマウス(Rip3-/- :Spink3SPINK1/-)を作製したところ、慢性炎症が抑制された。このことは、膵炎発症過程で見られる、オートファジー不全による細胞死がネクロプトーシスであることを示唆している。 以上の結果から、急性膵炎、慢性膵炎の発症にはオートファジーが深く関与しており、特に両者に共に出現する巨大なオートリソソーム内で、なぜ分解が抑制され、トリプシノーゲンの異所性活性化が生じるのかを明らかにする必要があると考えられた。
