プロトンチャネルノックアウトマウスを用いた摂食中枢機構の維持とその破綻の理解

Outline of Annual Research Achievements

昨年度に引き続き、野生型とVSOP/Hv1遺伝子欠損マウスの間で、NADPH oxidaseの局在に違いが認められるかを検証した。NADPH oxidase complexにおける細胞質サブユニットであるp67抗体を用いた免疫細胞化学を行った結果、野生型ミクログリアにおいてはp67とF-actinの局在に強い相関が見られたのに対し、VSOP/Hv1遺伝子欠損ミクログリアにおいてはそれが見られなかった。以上からVSOP/Hv1遺伝子欠損ミクログリアにおいてはF-actinの重合・脱重合異常によってNADPH oxidaseの細胞質ユニットの局在に変化が生じ、活性酸素産生の違いを生じていることが示唆された。
更にこのようなin vitroでのVSOP/Hv1による活性酸素産生調節機構が、in vivoにどのように反映されているかを明らかにするため、週齢の異なる野生型マウス及びVSOP/Hv1遺伝子欠損マウスについて酸化ストレスマーカーの発現量を調べた。その結果、生後1日、5日、3週間齢の VSOP/Hv1遺伝子欠損マウスでは酸化ストレスマーカーの発現量が低下していたのに対し、6か月齢では逆にこれが増大していることが明らかとなった。以上から、VSOP/Hv1による活性酸素産生機構はin vivoにおいては加齢依存的であることが示唆された。
また本年度の解析では、C57BL6/JのマウスからC57BL6/Nにバッククロスするにつれ肥満の表現型が小さくなっていたため、バックグランドによる影響が考えられた。しかしC57BL6/Nの4か月齢のマウスにおいてもVSOP/Hv1遺伝子欠損によって血中レプチン濃度が減少している傾向が認められ、摂食中枢の調節に関わっている可能性が考えられた。
