Outline of Annual Research Achievements

卵子は卵管内で精子と受精した後、着床前期胚として卵管内で発育し、胚盤胞となり着床直前に子宮内に移入する。子宮内膜管腔は、排卵直後はエストロゲンの作用によって鋸歯状となるが、その後の卵巣黄体化に伴い黄体ホルモンに暴露されることで、子宮内膜は適切な細胞増殖・分化の状態を獲得し、管腔はスリット状になる。子宮内膜上皮に着目すると、着床前～着床期にかけて起こる変化として細胞増殖の停止・細胞極性の減弱が挙げられ、いずれも子宮内膜の胚受容能獲得の必要条件である。これらの変化が起こることがその後の着床の成立には必須であり、この機構が障害されると着床しない。マウスin vivoの系を用いて、妊娠に必要な子宮内膜上皮管腔形成機構の解明を目指して研究を行った。本年度の研究により、（１）ホメオボックス遺伝子MSXの欠損マウス子宮では上皮の細胞極性低下し着床障害をきたすだけでなく、子宮が着床能を獲得する以前からNFkBの活性化が誘導され炎症応答により偽着床部位が認められること、（２）妊娠に必要な子宮内膜の上皮管腔の再生過程を観察する実験系として子宮脱細胞組織の移植マウスモデルを確立し、再生子宮により正常妊娠が維持できること、が明らかとなった。子宮内膜の上皮管腔形成の機構を検討した本研究により、着床を中心とする妊娠の成立・維持の仕組みの一端が明らかになった。
