Outline of Annual Research Achievements

植物表皮の通気口である気孔は、一対の孔辺細胞が膨張・収縮することにより開閉し、 光合成のためのガス交換と蒸散を調節する。気孔の存在は植物の生長と生存に必須なだけでなく、地球レベルの大気環境に影響を及ぼしている。気孔は、気孔系譜の幹細胞であるメリステモイド細胞から生まれ、 メリステモイド細胞の数は様々な外部環境に適合するよう巧妙に調節されている。本研究では、申請者が新規に所属することになった名古屋大学トランスフォーマティブ生命分子研究所(ITbM)の理念である「合成化学と生物学の融合による新奇化合物の創出」を目指し、メリステモイド細胞の維持と分化を司る内的因子、および合成化合物の探索と同定を目的とする。

平成27年度末までに、孔辺細胞特異的に強いGFP蛍光を発するシロイヌナズナレポーター植物を用いて現在までに約4200種類の化合物をスクリーニングした。その結果、気孔の数とメリステモイド細胞の数を増やす一群の新奇化合物が同定された。これらの中から効果が高く副作用が少ない候補をさらに選抜し、2つの化合物について詳細な解析を行った。その内の一つはメリステモイド細胞の増殖と気孔の分化の両方を著しく促進した。 驚く事に、この化合物は地上部および地下部のバイオマスを増大させた。この効果は、広く作物植物で認められたため、この化合物は気孔の分化とバイオマス生産の両方に共通するプロセスに関わることが示唆された。今後は、合成化学によるアナログ(類似化合物)のレパートリーを増やし、生理活性を担う化合物構造を狭めるとともに、標的因子の同定につなげたい。
