Outline of Annual Research Achievements

同種の雌雄は、次世代への遺伝情報の継承にあたって、協調的であると同時に対立的でもある。雌雄の行動上での駆け引きの背後には、神経ネットワークレベルでの雌雄差があり、その神経の性的二型はまた、遺伝子発現の性差に根差す。行動と神経の性差に対して最も大きな効果を有すると思われるのは、ショウジョウバエのfruitless (fru)遺伝子である。fru mRNAは雄の神経系でのみ翻訳されてFruタンパク質を生成する。Fruタンパク質はヒストン脱アセチル化酵素のHDAC1やヘテロクロマチンタンパク質1aと複合体を形成して染色体の標的部位に結合し、クロマチン修飾を介して一連の標的遺伝子群の転写を雄型の状態にすると考えられる。しかしこれまで、Fruの標的遺伝子は同定されていない。我々はとりわけ顕著な性的二型を示すmAL介在ニューロンに着目し、雄特異的突起の有無がrobo1遺伝子の機能に依存することを見出した。さらにゲルシフトアッセイ、レポーターアッセイによってrobo1遺伝子制御領域内にFru結合配列を同定し、robo1がFruの直接の標的であることを立証した。さらにrobo1遺伝子制御領域のFru結合配列だけを欠失した変異を作り出し、その変異体雄に固有の求愛行動の異常を見出した。この行動異常は、mAL特異的にrobo1をノックダウンすることで救済された。
