Outline of Annual Research Achievements

(1) 自家和合性の進化にみられる共通パターン
有性生殖のとくに自家不和合性反応では、雄と雌の利害対立とゲノム相関が重要な役割を果たす。新学術領域での共同研究の成果により、多くの自然種で繰り返して、雄特異性因子の変異で自家和合性が進化したことなど、適応進化での分子レベルでの法則性がみえてきた。これらについて、自殖についての総説を執筆した。現在、高山研究室との共同研究で、倍数体が自家和合性になるためにエピジェネティックな制御が重要である可能性の研究を進めている。
(2) 異質倍数体での両親ゲノムの組み合わせ
異質倍数体は、両親種のゲノムが組み合わさっており、ゲノム相関の解析のモデル系だが、これまで倍数体ゲノムの複雑さのためにほとんど解析が進んでいなかった。そこで、RNA-seq手法HomeoRoqを開発し、シロイヌナズナ属異質倍数体ミヤマハタザオの解析を進めている。ミヤマハタザオは片親ハクサンハタザオから重金属蓄積能を、もう片親のセイヨウミヤマハタザオから低温応答を受け継いで組み合わせてジェネラリストになることで分布域を拡大したことが示唆された。一方、これらのストレス応答遺伝子は異質倍数体では親の半分程度に下がっており、倍数体であることの代償(トレードオフ)のためにスペシャリストである親と競争しても勝てないことが示唆された。カルダミネ属植物C. amaraとC. hirsuta、およびそれらを両親種とする異質倍数体種C. flexuaosaにおいて、環境に応答して発現パターンの変わる遺伝子を網羅的に探索した。シロイヌナズナで孔辺細胞の大きさを制御するMPK12を調べたところ、C. amaraとC. hirsutaでリン酸化されると予測される部位に多型があり、C. flexuosaはどちらのホメオログも保持していた。またC. hirsutaタイプのMPK12がシロイヌナズナのmpk12変異体をより相補することがわかった。
