Outline of Annual Research Achievements

<最初期遺伝子を検索・同定>本年度はメダカの個体記憶を介した配偶者選択に関わる神経ネットーワークを包括的に同定する目的で、メダカ脳で最初期遺伝子を検索・同定した。方法はPTZ(GABA阻害剤)添加によって誘導される癲癇依存に発現が誘導される遺伝子群をマイクロアレイによって検索した。約15,000遺伝子について発現量を比較したところ、18種類の遺伝子が有為に発現上昇することを定量的RT-PCRで確認した。そのうち2つの遺伝子を用いて脳活動のマッピングを行ったところ、メダカメスがオスと長時間見ていた時に活性化している脳領域(終脳背側の一部)を同定した。
<脳領域特異的な条件的遺伝子操作法の確立>これまでに、メダカの条件的遺伝子操作法を確立し、成体脳において細胞系譜単位(単一の神経幹細胞から発生したニューロン群)を遺伝学的にラベルすることに成功している。さらに一部の細胞系譜単位は終脳背側ではブロック化していることを見出している(PLoS One 8:e66597,2013)。本年度は終脳背側の一つの細胞系譜単位が(1)で同定した活性化脳領域に対応することを見出した。
<個体記憶を介した行動を定量化する新規行動実験系の確立>本年度は新規な行動実験系を2つ確立し、以下の2点を示した。(1) メダカの三角関係(オス、オス、メス)において、オスはライバルオスに配偶者防衛行動を示すことで、メスがライバルオスを記憶することを妨害している(Frontier in Zoology in press) 。メダカメスはそばにいたオスを記憶して、配偶者として選択する傾向があるので、メスがライバルオスを選択することを妨害していると考えられる。(2) メスは排卵前には視覚記憶したオスに選択的に近づき行動を示すが、排卵後はオスに対する選択性がなくなる(Zoological Science in press)。
