Outline of Annual Research Achievements

神経細胞には活動の強弱によりシナプス強度や神経細胞そのものの興奮性が変化する「ホメオスタティック可塑性」という現象があることが知られている。光遺伝学によって海馬歯状回の神経細胞の興奮性を操作することでホメオスタティック可塑性を引き起こすことができれば、歯状回の成熟度についても変化を引き起こすことができると考えられる。本研究は、幼児性健忘が海馬歯状回の未成熟によって引き起こされており、海馬歯状回の成熟度が記憶の安定性を決める因子であるという仮説に基づいて行った。
本年度は、光遺伝学による歯状回特異的な神経活動操作が記憶の安定性にどのような効果を及ぼすか検討した。具体的には Cre-loxPシステムを用いて光感受性タンパク質であるチャネルロドプシン2(ChR2)を歯状回特異的に発現させてマウスの歯状回に in vivo で局所的に光刺激を与え、マウスに各種の記憶課題を行わせた。海馬歯状回を局所的に光刺激したマウスでは、いくつかの行動異常を確認することができた。しかしながら、記憶の安定性については明確な結果は得られていない。使用した刺激条件では海馬歯状回の成熟度が変化することは確認しているが、記憶の安定性に影響を及ぼすには成熟度変化の度合いが少ない可能性がある。
また、本年度はアデノ随伴ウィルスベクターの接種による発現システムの立ち上げを行った。アデノ随伴ウィルスベクターの接種により光感受性タンパク質の脳内での発現を確認することができた。
今後は、これまでに得られた成果を元に海馬歯状回の成熟度と記憶の安定性についてより詳細に検討を行う。
