Outline of Annual Research Achievements

分散効果(spacing effect)は様々な動物の記憶に共通した性質として、記憶の形成・維持など、記憶のダイナミクスにおいて重要である。分散効果は神経活動中に惹起された様々な変化とともに、トライアル間の“休憩”の重要性を意味するが、“休憩中”の詳細な分子・神経機構は不明である。最近、“休憩中”にROS(活性酸素種)が産生されること、ROSがNO(一酸化窒素)と相互作用して持続性のシグナル分子(8-ニトロ-cGMP)が生成されることが見いだされた。そこで本申請ではNO系が重要な役割を果たし、分散効果が観察されるマウスの小脳依存性運動学習に着目して、以下の仮説を検証した;「学習の分散効果には、トライアル間の休憩中に産生されるROSとNOの相互作用による持続性シグナルが寄与する」。
本年度は昨年度確立した視機性眼球応答(OKR)システムを利用して、OKR長期記憶における分散学習のトライアル間間隔(0-60分)を検討し、30分以上のトライアル間間隔が効率的な長期記憶形成に必要である事を見いだした。さらに、OKRを担当する部位である小脳片葉にROSのスカベンジャー(SOD, Catalase)を投与すると、短期記憶は正常であるが長期記憶が特異的に障害される事を見いだした。さらに、8-ニトロ-cGMPを模倣した阻害剤8-ニトロ-cGMPSも同様に長期記憶を特異的に障害する事を見いだした。ROS及びROS-NO系により生成される8-ニトロ-cGMPが長期記憶に関与する事が強く示唆された。
