Outline of Annual Research Achievements

ヒトの消化管には、体細胞の総数を上回る膨大な数の腸内常在菌が生息している。腸内細菌は、宿主の消化酵素では分解できない食物せんいなどを微生物発酵により分解し、終末代謝産物として様々な低分子化合物を産生している。宿主はこれらの『発酵代謝産物(以下、発酵産物)』をエネルギー源として利用している。腸内細菌によって供給される短鎖脂肪酸は腸上皮にとって最も重要な栄養源である。加えて、腸内細菌は糖・脂質代謝の補酵素となるビタミンB群を宿主に供給している。腸上皮は体内でもっとも細胞増殖の盛んな場所の一つであり、絶えず新生とアポトーシスを繰り返し、マウスでは約3日間で、ヒトでは約5日間で全体が入れ替わる素早い細胞代謝回転を行っている。腸上皮は発酵産物を有効に利用することでエネルギー(ATP)を獲得する以外に、ヒストンアセチル化やDNAメチル化の基質となるAcetyl-CoAやSAMなど生命素子の産生を増加させることが予想される。そこで本研究では、発酵産物を起点とした転写調節とエネルギー代謝の相互作用を明らかにする。
昨年度は発酵産物の供与によって変化する生命素子のメタボローム解析を実施した。その結果、腸内細菌定着により盲腸内容物の代謝物は劇的な変動を示し、それにより大腸上皮細胞内のエネルギーレベルが顕著に上昇する知見が得られた。今年度は大腸上皮細胞のトランスクリプト-ム解析を実施した。トランスクリプトームデータをGOに基づきfunctional enrichment analysisを実施したところ、無菌マウスに腸内細菌を定着後、早期にはcell cycleやproliferation関連遺伝子群の増加が顕著に観察された。続いて、後期には抗菌ペプチドを始めとする生体防御分子の発現が誘導されることから、細胞の成熟が進むことが示唆された。エピジェネティクス解析からこれらの生体防御分子の一部はヒストン修飾によって制御されていることが明らかとなった。
