Outline of Annual Research Achievements

Omics解析で明らかになった「共生器官に高発現する遺伝子」が、必ずしも共生成立において重要な役割を果たしているとは限らない。共生関連遺伝子の同定のためには、機能解析を行い検証する必要がある。本研究では、ホソヘリカメムシ-Burkholderiaモデル共生系を対象に、共生器官で発現する遺伝子を網羅的に調査し、発現遺伝子のRNAiスクリーニングおよび総合的な機能解析を行い、宿主ー共生細菌間相互作用に関わる宿主側の重要遺伝子を網羅的に同定することを目標に研究を行った。
前年度はホソヘリカメムシ共生器官において高発現している新規分泌タンパク質(システインリッチタンパク質)をターゲットに絞り、その発現パターンの調査やRNAi、組み換えタンパク質を用いたin vitro条件における機能解析を行った。今年度は主要なシステインリッチタンパク質や共生器官で高発現する消化酵素(カテプシン等)についてRNAiを試みたが明確な影響を検出することはできなかった。前年度までの研究によってin vitroとin vivoにおけるBurkholderia共生細菌の形状が異なる(共生時に菌体サイズが縮小し球状になる)ことが明らかとなっていたことから、本年はこれまでに得られていた共生細菌のRNAseqデータを精査し、共生細菌の形態に関わると考えられる遺伝子の抽出を行った。その結果、共生細菌はin vivo条件において細胞分裂関連の遺伝子(ftsZ, ftsI, dnaA, recAなど)が亢進していた一方、細胞壁合成に関わる遺伝子群(mreB, mrdA, mrdBなど)が大幅に発現抑制されていることが明らかとなった。これらの遺伝子の発現動態は、今後宿主の“共生細菌コントロール”因子をRNAiにより解明するための良い指標になると考えられる。
