Outline of Annual Research Achievements

共感性行動の基盤となる神経回路の起源を探るため、本年度はマウスにおいて情動伝染に重要であることが報告されている大脳皮質(帯状皮質)から背側視床、扁桃体への投射に関わる神経細胞について、爬虫類、鳥類胚を用いて解析を行った。脂溶性の軸索トレーサーと免疫組織科学的手法を組み合わせた解析の結果、爬虫類、鳥類の皮質相同領域から視床あるいは扁桃体に投射する神経細胞は爬虫類、鳥類にも存在するが、こうした神経細胞に発現する転写因子の発現様式は種間で大きく異なっていることが明らかとなった。特にBcl11b とSatb2 の2つの転写因子に着目すると、哺乳類ではこれらの転写因子が大脳皮質層ごとに排他的発現を示すのに対し、爬虫類や鳥類では1つの神経細胞が同時に2つの転写因子を発現していることが明らかとなった。この結果は、特定の神経回路を構築するための発生プログラムが種間で保存されていないことを示唆している。さらに、Bcl11b のゲノム配列の種間比較を行った結果、哺乳類のBcl11b ゲノム領域には 5'側と3' 側に転写抑制配列である Matrix Atachment Region (MAR) が存在するのに対し、鳥類では 3' 側に1つしか存在しないこと、個々の MARの転写活性をルシフェラーゼアッセイによって検討すると、鳥類や爬虫類の MARは転写抑制活性が哺乳類 MAR と比較して低いことが明らかとなった。これらの結果を総合して考えると、羊膜類(哺乳類、爬虫類、鳥類)の共通祖先の段階で、情動伝染に関わる神経回路はすでに確立されていたこと、しかしながらその回路を構築するための発生プログラムは種によって多様であること、哺乳類は大脳皮質層構造を獲得した段階で厳密な転写抑制機構を獲得し、これが哺乳類の情動伝染の神経回路の構築に必須に役割を持つようになったことが推測された。
