Outline of Annual Research Achievements

本年度は，前年度に開発した実験パラダイムを用いて，援助行動を行なっているラットにおいて活性化している脳領域について，神経活性化のマーカであるc-fos遺伝子の発現によって検討した．特に社会的行動に関連することが知られている，前部帯状皮質，扁桃体，前部島皮質領域の活性化について注目した．免疫組織化学的手法を用いて，c-fos遺伝子を発現している神経細胞の数を，援助行動が安定して出現するようになったラットと，援助行動を行なわせていない統制群のラットとの間で比較したが，注目した3領域については両者の違いが認められなかった．今回行なった実験では，安定した援助行動の出現の基準を，3日間連続して90秒以内に援助行動にあたるドア開け行動が出現することしたが，この基準が高すぎた可能性が考えられる．つまり，この時点では，既に援助行動が獲得済みであったため，注目した3領域の関与が弱まっていた可能性がある．そのため，今後は援助行動の出現初期の神経活性化を調べたいと考えている．また，3領域以外の領域についても検討する必要がある．
加えて，オキシトシンの投与が援助行動に与える影響について検討した．援助側の個体を，被援助個体とペアで飼育した場合と，単独飼育した場合とでオキシトシン投与が援助行動の出現に与える影響について検討した．その結果，オキシトシンの投与はペア飼育下ではあまり効果はなかったが，単独飼育条件においては，オキシトシンが投与されたラットは，生理食塩水を投与した統制群のラットよりも早く援助行動を出現させる傾向が認められた．オキシトシンと飼育環境の相互作用が示唆されるが，今回の実験では全身性にオキシトシンを投与したために，どの程度中枢神経系に投与の効果があったのか判断できない．今後は脳室内投与などの局所投与の効果を検討する必要がある．
