Outline of Annual Research Achievements

短期記憶・長期記憶などに分類されるように、生理学的に異なったプロセスを経て形成される記憶の成分比は、時間経過に伴って変化する。特に長期記憶は学習後一定時間経ってから形成され始めるなど、その形成過程には謎が多い。これを構成する神経細胞がいつ、どのように働くかの解明は、神経回路の発現する機能を理解するうえで不可欠である。我々は先行研究で、短期報酬記憶と長期報酬記憶は独立して形成されることを示した。さらに、長期報酬記憶を特異的に誘導するドーパミン細胞種を同定した。このドーパミン神経細胞は、昆虫の連合記憶中枢であるキノコ体の特定領域に入力する。本研究では、この領域からの出力神経が、ドーパミン入力神経に再び入力するというフィードバック回路を形成しているという仮説に基づき、その形態の詳細な解析を行った。
平成27年度は記憶の形成、固定化など様々なタイミングで強制的にフィードバック回路の機能阻害または活性化を行い、各時点におけるその重要性を調査した。この結果、これに関わる神経回路に機能不全を持つハエは長期記憶を形成できないことが分かった。すなわち、報酬情報がキノコ体のフィードバック回路によって保持され、安定した長期記憶の形成に重要な役割を担うということが証明された。この成果は、Ichinose et al., eLife, 2015に代表される4編の原著論文として報告した。
