| Outline of Annual Research Achievements |
本総括班研究では、包括的single-cell transcriptome(Nx1-seq)、遺伝子発現データ解析、エピゲノム解析、プロテオーム解析、リピドーム解析、マイクロバイオーム解析、遺伝子組換え動物作製を支援する炎症細胞社会解析センターを設置し、領域横断で使用可能な炎症細胞社会の解析プラットフォームを整備し、環境ストレス、ゲノム社会予防医学、脂質代謝生化学、循環器・肝臓・腎臓臨床医学、分子標的予防医学、計算生物学など多様なバックグラウンドを持つ研究者の連携を強力に促進することを目的としている。今年度は、次世代DNAシークエンサーを新規に導入し、包括的single-cell transcriptomeや遺伝子発現データ解析、エピゲノム解析の支援体制を整備した。計画研究【肺線維症の炎症細胞社会】の支援として、肺線維症モデルマウスから取得した病期の異なる肺細胞懸濁液について包括的single-cell transcriptomeを行い、正常、炎症極期、炎症終期における1000個以上のsingle-cell transcriptomeデータを取得し、計画班に提供した。また、エピゲノム解析、プロテオーム解析、リピドーム解析、マイクロバイオーム解析、遺伝子組換え動物作製などについて支援体制を構築した。総括班が主催した班会議において、各計画研究が分担する支援内容、費用負担などの取り決めを行うとともに、領域内での共同研究体制を構築した。また、ホームページを整備することで、本新学術領域研究ならびに計画研究の方向性を社会に発信し、さらに公開シンポジウムを通じて本学術領域の広報活動を行った。

Reason
計画通り次世代DNAシーケンサーを導入し、single-cell transcriptome支援の第1号を行った。この支援の成果が公開シンポジウムで発表されたことにより、領域全体でsingle-cell transcriptomeに関する意識の共有や、各計画研究の方向性が具体的になり、現在複数の計画班からそれぞれの実験モデルに関する支援要請を受けており、順次支援を行う予定である。

H30年度に引き続き、各計画研究、公募研究を支援する。領域の運営方針、研究支援内容は、年2回開催する班会議にて主に策定するが、必要に応じて随時委員会を開催し、領域代表（総括班研究代表者）の承認のもと、柔軟に対応する。①Nx1-seq:マイクロウェルデバイスを用い、固有のバーコードを持つ1細胞由来cDNA libraryを数千個の細胞について作成し、次世代シーケンサーを用いて各細胞の遺伝子発現情報を取得する。②遺伝子発現データ解析:発現変動遺伝子抽出、GO解析、クラスター解析、パスウェイ解析などを支援する。③エピゲノム解析：MSD-AFLP法などを用いたDNAメチル化解析、ChIP-seqなどによるヒストン修飾解析を支援する。④プロテオーム解析：蛍光標識2次元ディファレンスゲル電気泳動解析システムと質量分析装置（TOF-MS）を用いて標的タンパク質の同定を支援する。
⑤リピドーム解析：液体クロマトグラフィーと三連四重極型質量分析計（TripleQ）を組み合わせたLC-MS/MS、ならびに四重極飛行時間型質量分析計（Q-TOF）を用いて、リピドーム解析を支援する。⑥マイクロバイオーム解析：次世代シーケンサーを用いて、細菌叢の16S rRNA遺伝子配列に基づく網羅的解析を行い、腸内細菌叢遺伝子情報解析を支援する。⑦遺伝子組換え動物作製支援：CRISPR/Cas9などの遺伝子編集技術を用いて、ノックアウトマウス、レポーターマウスの作製を支援する。
