Outline of Annual Research Achievements

ノンターゲット解析から得られたMS/MSスペクトルのパターン認識、液体クロマトグラフィーにおける保持時間、イオンモビリティにおけるCCSの情報全てを統合することで、約8,000種のリポクオリティを捉える情報処理技術の開発に成功し論文発表した。この過程で、従来の複合脂質クラスを大きく超える117クラスの特徴的なMS/MSスペクトルを明らかにした。このように実測データに基づくMSライブラリーの構築を進め、幅広くリポクオリティの違いを識別する解析システムの構築が順調に進んでいる。これらを用いて、アラキドン酸、EPA、DHAなど異なる脂肪酸を摂取した後の代謝動態や分布の包括的解析、精子の正常発達に必要な脂肪酸環境（DHA/DPAが豊富）をつくる仕組み、腸内細菌のリノール酸代謝による肥満改善効果（公募班木村と共同）、母体腸内細菌由来の短鎖脂肪酸による子の肥満改善効果（公募班木村・長谷と共同）などについて論文発表した。さらにLipidomics Standards Initiative (LSI)の主要メンバーとして、リピドミクス解析データのクオリティ指標について国際ガイドラインを提唱するオピニオン論文を共同発表した。
瀬藤グループは、EPAやDHAを投与した動脈硬化モデルマウスの弁部および弓部を質量顕微鏡法および免疫染色法で解析し、動脈硬化プラークに対するEPAとDHAの作用の違いを組織学的に明らかにした。質量顕微鏡法で得られたデータを機械学習により自動解析する事で、マウス脳の分子分布パターンを大別し、小脳皮質特異的に分布する脂質群を発見した。認知症モデルマウスにグリーンナッツオイルを投与する事で、認知症の改善効果を見出し、その時の脳局所における脂肪酸分布の変化を質量顕微鏡法により明らかにした。
