Outline of Annual Research Achievements

哺乳類培養細胞でのイノシトールリン脂質の動態を解析する効果的な分子ツールとしてこれまでに世界的に用いられてきた脊椎動物ゼブラフィッシュ由来Voltage-sensing phosphatase(Dr-VSP)について、前年度までにホヤ由来VSP(Ci-VSP)での詳細な解析から見出したホスファターゼドメイン中のhydrophobic spineの膜脂質との相互作用候補アミノ酸部位を改変することにより、電位依存性酵素活性を向上させることによる分子ツールの改良が可能かを、主に発現系細胞HEK293T細胞での電気生理学的解析、蛍光顕微鏡をもちいた発現解析によって検討した。
これまでの脂質のリポクオリティを認識する分子機序としてCi-VSPのhydrophobic spineのL284に対応するL223をFに変異させた変異体は電位依存的酵素活性を1.7倍増加させた(Kir2.1活性での評価)。更に細胞膜への限局を向上させるため、これまでの解析から細胞膜へのターゲットを向上させることが知られていたCi-VSPのN末端側細胞内領域とのキメラを作成した(CiDr-VSP-mChe-L223F)(表面での限局を確認するため蛍光タンパク質も融合させた)。これにより野生型のDr-VSPよりも2.1倍から2.8倍に電位依存的酵素活性を向上させることに成功した。また、研究分担者の中川のチームによる解析によって、様々なイノシトールリン脂質を基質としたVSPの共結晶を作製し高分解能での構造情報を得ることに成功した。

Reason

これまでに明らかにしてきたVSPの膜との相互作用部位hydrophobic spineが、ホヤ由来VSPだけでなく脊椎動物のVSPについても保存されていることが明らかになり、さらにこの部位のアミノ酸変異により、イノシトールリン脂質のレベルを短時間で変化させる分子ツールとしての性質を向上させることに成功した。VSPのX線結晶解析についても、基質との共結晶が複数得られており、結合部位の構造基盤が明らかになりつつある。

Strategy for Future Research Activity

VSPについてはリポクオリティ認識機構の原理を深く知るため、Ci-VSPについて、これまでに確立してきた発現系細胞での非天然アミノ酸ANAPを用いたVoltage clamp fluorometry法を更に工夫し、側鎖間での相互作用を解析する。これまでに電位依存的なスペクトルの変化の計測が可能であったH182Anapのコンストラクトを用いて、更に解像度の高いシグナルを得られるかを検討する。またこのシグナルの分子機構について、他の部位に2つ目の変異を導入するなどによる解析をおこなって明らかにする。VSPの構造については、これまでに得てきた様々な基質との共結晶構造について更に分解能を上げられるかを検討する(研究分担者中川)。
