Outline of Annual Research Achievements

最終年度となる2019年度は、これまでに本プロジェクトで新たに提案した明るいプローブを用いた、ナノプローブの生体内における未知の挙動の描出、昨年度までに構築したCT原理による三次元イメージングに時間軸を合わせた蛍光寿命三次元分布描出システムの構築に取り組むことにした。その結果、シートレーザー原理では顕微鏡とは異なり、マクロスケールではライトシートのコントロールが困難であることがわかった。そこで、CT原理に注力を映した結果、被写体中の3次元のTime Gated Imaging(TGI)に成功し、3次元での蛍光寿命の分布を描出することができた。CTイメージングはこれまでに引き続き横田班との連携により進めた。さらに蛍光寿命を温度に変換することにより蛍光寿命から推定される被写体内の温度の描出が可能になった。
ナノプローブについては特に色素内包プローブにおいて描出された、血管到達速度、胆汁排泄経路に注目し、生体内挙動を明らかにすることを狙いとした。特に、DSPE-PEGを用いて作製した疎水コア見せるにおいては、血中滞留後に肝臓に到達したプローブが胆汁排泄経路にのって小腸へ運ばれ、腸内で蛍光を放つことが明らかになった。
透明化試薬の実証実験は根本班との連携により脳神経脳イメージングにおける短時間での透明化実証を行った。この結果、従来の試薬よりもはるかに高速な1時間程度での透明化を行ったマウスの脳のイメージングを実現することができた。さらに、最終年度に当たり、このプロジェクトを通して得られたSBWにおける蛍光イメージングの新たな境地の開拓の総まとめとして、世界的に見てもまとまった知見のない生体光イメージングにおける透明性を詳説した成書の執筆を開始した。これについては2020年夏ごろの出版を目指している。
