Outline of Annual Research Achievements

環境刺激を受けるとクロマチンにエピジェネティックな変化が引き起こされる。その変化を制御する分子メカニズムを、植物のDNA損傷ストレス感受性を利用することで、ヒストン修飾やクロマチンリモデリングに関与する植物のクロマチンの動態制御因子を同定し、その機能を明らかにした。
1)クロマチンの核内配置を制御メカニズム
シロイヌナズナの細胞核では、ヘテロクロマチンであるセントロメア・クロマチンが核膜周縁部に分散配置する。この分散配置に関与している複合体を同定した。一方、核質側ではコンデンシンを介してセントロメア・クロマチンと相互作用する。各因子の変異体のHi-C解析を進めて、この複合体が全ゲノムレベルでDNA間の相互作用に影響を与えることがわかった。この複合体の変異体はDNA損傷に高感受性となることから、植物のクロマチン配置は環境刺激に対する応答メカニズムに関与していることが示唆された。
2)植物の器官再生におけるエピジェネティック・プライミング
エピジェネティック・プライミングは、将来の再生や分化に備えた待機状態において、エピジェネティクス変化やRNA polIIのpoisingなどが起こる現象である。シュートが再生されないヒストン脱メチル化酵素の変異体ldl3を同定し、ChIP-seqとRNA-seqの統合解析の結果、LDL3はヒストンH3K4のメリル残基を脱メチル化することがわかった。さらに、LDL3は細胞記憶の消去に関与しておらず、シュートの再生に必要な遺伝子群のH3K4をあらかじめ脱メチル化していた。つまり、シュート遺伝子群の発現をエピジェネティック・プライミングにより発現スタンバイ状態にしている仕組みが植物にあることがわかった。
