Outline of Annual Research Achievements

インフルエンザウイルスは、人間界では病原微生物として認識されているが、自然宿主であるカモなどの野生の水禽では、感染しても病気を発症することはない。 本研究では、インフルエンザウイルスと水禽との共生の自然界における意義を理解するために、１）インフルエンザウイルスとの共生が水禽の腸内微生物叢や生理機能に及ぼす影響の解析、２）インフルエンザウイルスと水禽との共生メカニズムの解析、および ３）野生動物におけるバイローム解析による新規ウイルスの探索を行っている。
インフルエンザウイルスに感染した野生の水禽が病気を発症することはないが、感染個体においてどのような宿主応答が起きているかはよく分かっていない。そこで、本研究において、鳥インフルエンザウイルスを感染させたマガモにおける宿主応答を調べたところ、感染初期には抗ウイルス活性およびインターフェロン応答に関わる遺伝子群の発現上昇が見られた。それに対して、感染後期には、B細胞や免疫グロブリンが関与する免疫応答に関わる遺伝子群の発現が上昇していることが分かった。以上の結果から、鳥インフルエンザウイルスを感染させたマガモの腸管では、感染初期(1,3日目)に自然免疫応答が、そして感染後期（6,9日目）には獲得免疫応答が誘導されていることが明らかとなった。
野生動物におけるバイローム解析については、鹿児島県に飛来するツルの糞便サンプルのメタゲノム解析によって、新規のツルアデノウイルスを同定した。またアフリカ・シエラレオネで採取したコウモリサンプルの解析を進めている。さらに、令和元年度は、南米・ブラジルにおける野生動物のサンプリングのため、新たに共同研究を開始した。

Reason

本研究では、インフルエンザウイルスと水禽との共生が、水禽の腸内環境に与える影響を調べるため、腸内微生物叢のメタゲノム解析を行う。また、本ウイルスとの共生が、水禽の免疫系や生命活動に及ぼす影響を明らかにする。さらに、国内外の野生動物におけるバイローム解析によって、新規ウイルスの探索を試みる。カモを用いた鳥インフルエンザウイルスの感染実験を行ない、感染個体の腸における宿主応答解析を行ったところ、感染初期には抗ウイルス活性およびインターフェロン応答に関わる遺伝子群の発現上昇が、そして、感染後期には、B細胞や免疫グロブリンが関与する免疫応答に関わる遺伝子群の発現が上昇していることが明らかとなった。さらに野生動物のバイローム解析用に、アフリカ・シエラレオネのコウモリの臓器から抽出したRNAを用いて、次世代シークエンスを行い、バイローム解析を行っている。また鹿児島県に飛来するツルから採取したサンプルのメタゲノム解析の結果、新規ツルアデノウイルスを同定することができた。以上の結果が得られたことから、本研究は、おおむね順調に進展していると考えられる。

Strategy for Future Research Activity

今後も引き続き、国内外における水禽や野生動物から採取したサンプルの解析データを用いて、新規ウイルスの探索を試みる。具体的には、公募班の中川先生（東海大）との共同研究によって、シエラレオネとブラジルで採取したコウモリの臓器サンプルから抽出したRNAの次世代シークエンスデータを用いてバイローム解析を行う。さらに、鹿児島県で採取したツルの糞便データを用いたバイローム解析を行う。また、感染実験したカモから採取した糞便サンプルを用いて、腸内微生物叢のメタゲノム解析を行う。インフルエンザウイルスを保有している個体と保有していない個体における腸内微生物叢の違いを調べ、宿主応答などとの関連性について調べる。
