Outline of Annual Research Achievements

作物に感染し甚大な被害をもたらす糸状菌には、極めて多様なウイルスが高率に感染している。申請者は、既成のウイルス概念があてはまらない「裸性」・「宿借・宿主性」というユニークな生活環を持つ菌類ウイルスを果樹の病原糸状菌から発見した。本課題では、①ハダカウイルスの裸性(裸のRNAで感染組織で存在)、ヤドカリウイルス(YkV1)とヤドヌシウイルス(YnV1)の宿借・宿主性(ウイルス間の新しい相利共生関係)の証明とその分子基盤、②宿主菌生存圏の高次生態系(ウイルス・菌・植物)での前記ウイルスの役割(菌宿主の植物宿主での生育・生存適応能)の解明、さらに、③ネオ・ウイルスライフスタイル(裸性・宿借・宿主性)の普遍性を証明、を目指した。複数のFusarium属菌からハダカウイルス(HadV1)2系統を発見し、詳細に解析した。HadV1は10~11本の分節一本鎖RNAゲノムを持つこと、無性胞子と通じた垂直伝搬の効率は感染菌糸コロニーが古くなる程低下する、ゲノムRNA複製型dsRNAは共に感染菌糸破砕液中でRNAase感受性を示す(粒子とは異なり、裸状のRNA?)、HadV1は宿主菌に不顕性感染し平和的に共存していること、を明にした。YnV1にヘテロ粒子化されたYkV1が、自身のRNA合成酵素を利用して複製し、RNA合成酵素の活性化には2A様ペプチドを介したリボソームスキップ機構による翻訳が必須である、YnV1+YkV1はしゅくしゅきんの病原力衰退を惹起すること、を証明した。さらに、新規のヤドカリウイルス、ヤドヌシウイルスをスペイン産白紋羽病菌、バングラデシュ産糸状菌から、それぞれ発見した。これは、ヤドヌシ/ヤドカリウイルス間のパートナーシップ多様性の解明、ネオライフスタイルの普遍性の証明につながった。以上のように、ネオ・ライフスタイル(宿借・宿主性)の分子解剖・確立につながった。
