Outline of Annual Research Achievements

本研究課題では、生体内には存在し得ない人工生合成系を試験管内で構築すること、さらにこれらを用いて任意の生物活性をもつ天然物様化合物を創製することにより、新機軸の生物活性分子開発に資する『擬天然物合成生物学』の基盤を確立する。具体的には、改変翻訳系と異種由来のペプチド修飾酵素を複数組み合わせることで、各種主鎖修飾骨格を有する擬天然物ペプチドの試験管内リデザイン生合成系の創製を行う。さらに、この系を用いて、大規模な多様性(一兆種類以上)をもつ擬天然物のランダム配列ライブラリーを構築し、当該ライブラリーから新規生物活性擬天然物の探索系を構築することで医療に資する化合物の発見も視野に入れた研究を進める。
当該年度の研究では、チオペプチドと呼ばれる高度に修飾された骨格を有する天然物ペプチドの生合成クラスターの一つを試験管内で再構成することに成功した。また、この試験管内生合成系が高い基質許容性を示すことを実証した上で、多数の人工チオペプチドアナログの生産にも成功した。さらに、高い基質許容性を示す可能性の高いトリプトファンプレニル化酵素を組みこんだ試験管内リデザイン生合成系の構築にも別途成功した。これら二種の試験管内リデザイン生合成系は当初の計画では盛り込んでいなかった成果であり、研究課題全体として、多種多様な骨格の擬天然物ペプチドを生産するための幅広い生合成ツールを用意することに成功し、今後の擬天然物ライブラリー構築及び生物活性擬天然物探索の下地が整いつつある。

Reason

当初予定していた酵素の発現精製が不調であったため、軽微な計画変更を行ったが、別の菌株由来のホモログ酵素を使用することでこの問題を解決しており、研究課題の進捗に大きな影響はない。一方、当初の計画では想定していなかった知見を基に、チオペプチド及びプレニル化トリプトファン含有ペプチドの生産を行う試験管内リデザイン生合成系の構築に成功し、全体の研究計画通りもしくはそれ以上に順調な成果を上げていると自負している。

Strategy for Future Research Activity

これまでの研究により構築に成功した各種試験管内リデザイン生合成系について、そのさらなる実証実験と高度化を進める。さらに、これらの生合成系を用 いて、擬天然物ペプチドのライブラリーの構築とその試験管内選択系の構築・実証も行う計画である。
