Outline of Final Research Achievements

In this study, we have studied GPI modification of GPI-anchored proteins, phosphorylation of extracellular domains of membrane proteins, and special glycosylation of dystroglycans to elucidate the localization of the zones, the molecular mechanism of zone formation, and their physiological functions.
We found that some of the GPI-modified zones are formed in the nuclear envelope and adjacent endoplasmic reticulum by lamin, and that the zones are important for biological functions. We also found that phosphorylation zones are formed in a subset of Golgi complexes by the actin regulator twf. In addition, we found that a group of molecules involved in modification of dystroglycan form a complex, and that abnormal formation of the complex causes symptoms such as muscular dystrophy, cardiomyopathy, and lissencephaly.

Academic Significance and Societal Importance of the Research Achievements

分泌および膜蛋白質の翻訳後修飾を司る選別輸送ゾーンのいくつかについて、その局在、分子基盤、生理機能を明らかにすることができ、オルガネラゾーンの分野を大きく前進させることができた。学術的には日本初の新しいコンセプトを発信することができたと考えている。社会的意義については、ジストログリカンの修飾ゾーンの異常によって、筋ジストロフィーや心筋症、滑脳症といった症状が引き起こされることを見出し、それらの疾患の理解が進んだと考える。また、その治療法の可能性についても動物実験で良好な結果が得られたのは意義ある結果だと言える。
