Research Abstract

本研究課題では、極めて高い遺伝的多様性を維持しているマラリア原虫をモデルとして、寄生適応と病原性に関わる寄生体の遺伝的多様性の分子基盤を明らかにすることを目的とする。本年度は以下の研究成果を得た。
1.我々が展開しているマラリアワクチン実用開発において使用している抗原遺伝子SERA5についてフィールドサンプルより多型解析を行った。その結果、8mer反復配列の重複数に多型性が生じている事が明らかとなった。また、種々の8mer反復配列を組み込んだ改良型SE36蛋白質を発現・精製し、マウスに免疫することによって、SE36マラリアワクチンの改良に道を開いた。
2.熱帯熱マラリアに次ぎ流行をしている三日熱マラリアの高度の多重感染について、昨年度の解析に加え4個のマイクロサテライトを解析し、確実なものとした。
3.マラリア原虫の宿主域は広い(哺乳類、鳥類、爬虫類)が、各原虫種は特定の動物だけに感染する。この二つの事実はマラリア原虫と宿主との密接な関係を示唆する。この関係の把握を目的に、ミトコンドリアゲノムを用いてマラリア原虫と宿主の進化の比較を行った。その結果、現生マラリア原虫の起源(約3,800-2,500年前)において急速な多様化が起き、その多様化は宿主転換によって生じたことを見いだした。
4.msp1に働く正の選択を三日熱マラリア原虫(P.vivax)及び近縁サルマラリア原虫9種を対象に進化集団遺伝学的に解析した。その結果、P.vivaxを含む数種に共通(=種分岐前)の枝において正の選択が働いたこと、また、P.vivaxを含む数種では種を超えたアミノ酸多型の共有が存在することを見いだした。一方、種内多型を調べた結果、種によって正の選択の働く領域とその強度が異なっていた。
