| Research Abstract |
本研究では、中枢性体液調節系の主軸であるバゾプレッシン系を中心に研究を進めている。(1)ラット脳スライス標本を用いて、パッチクランプ法によりバゾプレッシン産生細胞が局在する視索上核の神経分泌ニューロンへの興奮性および抑制性シナプス入力に対する浸透圧変化とペプチド(アンギオテンシンII)の影響を検討した。その結果、高浸透圧刺激およびアンギオテンシンIIによって興奮性後シナプス電流が増加すること、この増加反応はTRPVブロッカーで消失することが明らかとなった。一方、抑制性後シナプス電流は高浸透圧刺激にも低浸透圧刺激にも影響されないが、高浸透圧状態→低浸透圧状態に変化した場合には増加反応が見られた。この反応に、BDNFを介したGABA_<A>受容体のインターナリゼーションが関与している可能性を見出した。(2)バゾプレッシンーeGFP(緑色蛍光タンパク)トランスジェニックラットの視索上核から単離したeGFP陽性ニューロンからパッチクランプ法により電流記録を行い、浸透圧変化、pH変化、種々のペプチドに対する反応性について検討した。(3)ラット視床下部室傍核、視索上核および下垂体後葉におけるTRPファミリーの存在の有無について免疫組織化学的染色法、RT-PCR法、in situハイブリダイゼーション法を用いて網羅的解析を行った。(4)無麻酔下のラット脳室内にアンジオテンシンIIを投与すると著明な飲水行動が惹起される。この反応が、グレリンもしくはTRPV4アゴニストである4αPDDの前投与によって抑制されることを見出した。一方、hypovolemiaによって惹起される飲水行動はグレリンの前投与では抑制されたが4αPDDの前投与では抑制されなかった。
