Outline of Annual Research Achievements

生命環境では、様々なタンパク質が特異な集合構造を形成し、自発的に機動する分子機関が構築されている。本研究では、代表者が開発した、T4ファージの構成部品であるgp5タンパク質を基盤とし、新しい分子機関の合成をめざした。具体的には、巨大分子集合体がさらに集合化して構造体として組み上がるためのタンパク質針精密設計として、gp5の結晶構造やHSAFMの実験結果と分子動力学計算を元に、分子機関としての機能を精密に制御したタンパク質針を実現した。二次元表面の上でgp5の精密な集積を実現するため、異方的な集合構造をとるたんぱく質改変を施した。その結果、高速AFMの実験結果を数理モデルを用いた解析により理解することによって、理論計算からモデルを構築し、新たな集合体形成のメカニズム解明へとつなげることができた。さらに、これらの結果を踏まえ、タンパク質の異方的な二次元表面への集積とその集団運動の制御に向けた精密分子設計の指針ができた。
