Grant-in-Aid for Scientific Research on Priority Areas
Arabidopsis ASYMMETRIC LEAVES2 (AS2) and AS1 genes control leaf morphogenesis from the upstream of its developmental pathway, but the regulatory framework for the morphogenesis remained to be demonstrated. Our group has investigated a role of these genes in adaxial-abaxial establishment and formation of symmetric flat leaves. We have demonstrated that AS2 and AS1 proteins form a complex, which plays a key role for the development of leaves with such characteristic architectures by repressing expression of abaxial gene ETTIN/ARF3 in presumptive adaxial domain in leaf primordia. The repression mechanism includes systems including binding of the complex to the ETTIN promoter region, enhancement of the level of miR390 that is involved in degradation of ETTIN mRNA, and epigenetic silencing of ETTIN.
