Research Abstract

1, オーキシンの極性を持った分布の構築機構の解析;オーキシンの偏差分布に関連する因子として、オーキシン細胞外排出タンパク質PIN、その調節に関与するキナーゼPIDが知られている。本研究は、pidのエンハンサーとしてMAB2とMAB4を単離し解析している。重篤な形態異本常を示すMAB4ファミリー遺伝子群多重変異体mab4 mel1 mel2においてPIN1タンパク質の細胞膜上の存在量が著しく低下し、同時に極性分布も弱まった。同様の傾向が重力屈性に異常を示すmel1 mel2 mel3 mel4四重変異体におけるPIN2タンパク質の細胞内局在にもみられた。MAB2は基本的転写共役因子メディエーターの制御複合体(CDK8サブユニット)を構成するAtMed13をコードしていた。meb2の表現型の解析から、MAB2はシロイヌナズナにおいて、胚発生過程でオーキシン応答に依存した転写を制御している可能性が示唆された。
2, 茎頂メリステム形成形成と維持の分子機構の解明;茎頂分裂組織(SAM)と腋生分裂組織(AM)に異常を示すuni-1D変異体の解析を通じて、分裂組織の全く新しい制御機構の発見を目指した研究を行ってきた。本年度の成果は以下の通りである。ERECTA (ER)受容体キナーゼの機能欠損によりuni-1D変異体のSAMの異常が回復した。また、uni-1D変異体のSAMの異常にはSAM外部でのERの機能が重要であった。さらに、ERファミリー因子群の機能をすべて欠損させると、uni-1D変異体のAMの異常もまた抑圧された。これらを踏まえた解析の結果、野生型背景においても、分裂組織外でのERファミリーの機能が分裂組織の制御に関わることが示唆された。
