| Research Abstract |
本年度は以下の3テーマについて解析を行った。1) TDIF-TDR(TDR receptor)の下流のシグナルカスケードの解析、2) 原生木部形成に関与する新規CLEペプチドの機能解析、3) 2つの木部細胞-木部繊維と道管-の分化制御の解析。
1) TDR受容体の下流のシグナルカスケードを解析した。まず、WUSのホモローグであるWOXファミリーの遺伝子の発現がTDIFで誘導されるかどうかを調べた。WOX遺伝子のうち、WOX4遺伝子のみがTDIFにより1時間以内に発現の促進がおこった。WOX4の機能欠損変異体では、前形成層細胞数の減少が見られた。複数の他の結果と合わせて、WOX4はTDRの下流で前形成層細胞の分裂を促進する転写因子として機能していることが明らかとなった。2) TDIF以外のCLEペプチドが維管束形成の制御に関与する可能性を、合成ペプチドの投与実験により検討した。その結果、複数のCLEペプチドが根における原生木部分化を阻害することが示された。次に、木部分化阻害の分子機構を知る目的で、CLEペプチドを投与した植物体の根を用いてマイクロアレイを行った。その結果、CLEペプチドとサイトカイニンとの関連性が明らかになった。サイトカインシグナルの変異体を用いた解析から、原生木部道管の形成において、CLEペプチドシグナルとサイトカイニンシグナルがクロストークしていることが明らかとなった。3) 木部繊維細胞のマスター遺伝子NST1と後生木部道管のマスター遺伝子VND6をエストロゲン誘導型にしてシロイヌナズナ培養細胞に導入し、形質転換ラインを作成した。これらラインを用いてマイクロアレイを行い、マスター遺伝子により直接支配される遺伝子群を同定した。
