Outline of Annual Research Achievements

本課題では、①生命起動メカニズムの解明、②卵活性化機構の解明をメインテーマとし、さらに③精子エピゲノム獲得機構の解明をサブテーマ として研究を進めている。
①生命起動メカニズムの解明:2021年度も引き続き、卵・卵巣特異的に発現する遺伝子について、CRISPR/Cas9 によりKOマウスを作製し、交配テストを実施した。11遺伝子のKOマウス系統を作って解析したところ、雌の妊孕性が低下した遺伝子が2つ、低下が認められなかった遺伝子が9つであった。
②卵活性化機構の解明:受精卵は受精刺激により活性化される。PLCZ1 以外に受精・活性化に影響する因子を探索して機能解析している中で、卵との融合不全を示す1700029I15Rik KOマウスを新規に樹立した。1700029I15Rikにコードされるタンパク質は小胞体において糖鎖修飾を認識し、受精融合に働く精子膜タンパク質群の品質管理を行っていることを明らかにして論文報告した。このほか、mRNAワクチンで注目されているLipid NanoParticle (LNP) に着目し、レンチウイルスベクターなどと異なり、LNPはアデノ随伴ウイルスベクターと同様に、透明帯を通過して受精卵に核酸を導入できることを見出した。卵活性化メカニズム解析およびゲノム編集マウス作製ツールとしての応用可能性について検討を始めている。
③精子エピゲノム獲得機構の解明:精巣上体の成熟およびその結果として精子受精能力を成熟させるルミクリンに着目して解析を進め、NELL2と結合して働く因子を発見NICOLと命名し、KOマウスを作製して分子機能を明らかにして論文報告した。

Reason

卵および卵巣特異的に発現する遺伝子群についてKOマウスの作製と解析は順調に進んでいる。22年度は9つの因子が必須でないことを明らかにすると同時に、必須因子も2つ同定できた。2023年度は解析に重点を移す。そのほか、40年来の謎であった精巣由来因子による精巣上体の成熟と精子受精能力獲得を司るルミクリン機構の解明も着実に進んでいる。

Strategy for Future Research Activity

引き続き、篠原の得意とするGS細胞、伊川が得意とするゲノム編集マウスのメリットを活用し、in vitro, in vivoの両観点から受精および卵活性化・受精後発生に寄与する因子の同定と機能解析を進める。また新規開発したLNPを用いた受精卵mRNA導入法の活用を進める。
