Outline of Annual Research Achievements

①これまでの本研究課題にて、ポンプープローブ時分割XFEL結晶構造解析法を確立し、多様な光駆動型膜タンパク質の構造ダイナミクスを解明してきた。しかしながら、生体内の大部分のタンパク質は光動作性ではないので、この方法はごく一部のタンパク質にしか適用できないことになる。この弱点を補完するために、B01清中班と連携して、天然では光動作性を示さないGPCR(ヒトアデノシンA2a受容体)に対して光異性化するアゾベンゼン修飾リガンド (基底状態ではpartial agonist)を開発した。当該化合物をアゴニスト安定化A2a変異体に結合させた複合体を脂質キュービック相結晶化法により結晶化し、静止X線結晶構造を解明した。並行してリガンドの構造改変も進めた。
②新規光駆動型膜タンパク質の自然界からの発見、構造ベースでの改変等を目的として、陸域の特殊な環境以外に生息する微生物が集光アンテナを備えたロドプシンを持つのかという点に着目して淡水湖および海洋に生息する環境微生物の調査を行った。その結果、レチナール色素に加えてカロテノイド色素の一種であるキサントフィル(ゼアキサンチンやルテイン)も結合するロドプシンを持つことを発見した。さらに、このキサントロドプシンが、なぜ4-ケト環構造を持たないゼアキサンチンを集光アンテナとして使うことができるのかをクライオ電子顕微鏡を用いた単粒子構造解析により構造学的に解明した。本研究により、水圏生態系においてロドプシンは集光アンテナを駆使し、従来の試算を上回る量の光エネルギーを受容することが示唆された。
