Research Abstract

本研究では、生殖細胞系列が獲得するユニークなエピゲノム情報とその獲得機構の解明、およびこれらの過程を試験管内で正確に再構成することを目的とし、1)生殖細胞形成に必須な転写制御因子Blimp1及びPrdml4が形成する蛋白質複合体の同定、2)生殖細胞誘導シグナル機構の同定と試験管内生殖細胞誘導系の開発、を推進する。
本年度までの研究で、転写制御因子Prdml4が生殖細胞形成過程に必須の役割を果たし、始原生殖細胞(Primordial Germ Cells:PGCs)における潜在的多能性の再獲得とエピゲノムリプログラミングに重要であることを示した(Yamaji M, et al. Nature Genetics, 40,1016-1022, 2008)。またPrdm14の作用機序を明らかにする目的でPrdm14を過剰発現する細胞株を樹立し(ES及びHela)、それらからPrdml4が形成する蛋白質複合体を抽出・精製し、現在複合体構成蛋白質の配列決定を行った。またPGCsが胚体外胚葉(エピブラスト)から誘導されるシグナル機構の解明に成功し、エピブラストから無血清条件下で選択的にPGC様細胞を誘導、それらがPGCマーカーを順序よく発現しエピゲノムリプログラミングを起こし、移植により精子形成及び正常な胎仔形成に寄与することを示した(Ohinata Y, et al. Cell, accepted in principle)。これらの研究成果を受け、今後Blimp 1及びPrdml4の作用機序を明らかにすること、ES細胞より選択的にPGC様細胞を誘導することが大きな目的となる。
