Research Abstract

哺乳類生殖系列細胞は、初期胚に存在する多能性細胞から派生し、ゲノムの再プログラム化というエピジェネテイックなゲノム再編や減数分裂を経た後に、配偶子となり次世代へと遺伝情報を伝えていく。本研究では、この細胞系譜に属する内部細胞塊、エピブラストなどの胚性未分化細胞、始原生殖細胞(PGC)の発生過程における全遺伝子の発現プロファイル動態の解析や、細胞核構造の変化やX染色体活性を指標としたゲノム再プログラム化過程の解析を行う。また、発生プログラムの転換点の詳細な解析を通じて、そのエピジェネティック制御機構の解明を試みる。変異体等を利用し、発生過程で特徴的に変動する遺伝子群の機能解析を行ない、さらに得られた知見を統合してin vitroでの発生イベントの再現を行っていくことを目的とする。
