Research Abstract

体細胞クローン動物の成功率は現在でも数%しかなく、エピゲノムによる異常がほぼすべてのクローン動物に生じてしまう。その原因は卵子による初期化が不完全であるためだと考えられているが、初期化機構そのものがまったくわかっていない。そこで本研究では、核移植技術を様々な条件で行い、DNA及びヒストンの修飾変化、卵子細胞質の効果、インプリント遺伝子の発現変化、およびマウスの遺伝的バックグランドの影響などについて検討する。最終的に卵子による核の初期化機構の解明を目指すと同時に初期化の促進方法を開発し、体細胞核の完全初期化および異常が生じない健全なクローン個体を高率に作出することを目指している。
