Research Abstract

二重鎖DNAを表層に密生させた高分子ミセル(DNA担持ナノ粒子)のコロイド安定性と電気泳動移動度が、DNA自由末端側の塩基対構造に明敏に応答することを発見した。自由末端に一塩基ミスマッチが存在すると、完全相補の場合と比べて、コロイド安定性と電気泳動移動度が著しく増大する。これは、DNA密生相とバルクとの界面における分子構造のわずかな変化がマクロでダイナミックな現象を誘起していることを意味する。本研究は、この特異な界面現象のメカニズムを分子レベルで解明し、新しいバイオ分析デバイスへ応用することを目的とする。具体的には、(1)新規高分子-DNAコンジュゲートの精密重合、(2)DNA担持ナノ粒子が示す非架橋型凝集の構造科学的観点からの解明、(3)新規バイオセンサー開発、に関する研究テーマの推進をした。
