Research Abstract

タンパク質中のアミノ酸変異は、タンパク質の揺らぎ研究において有効な研究手段である。しかし、従来の研究では使用できるアミノ酸は天然の20種類に限られていたために、精密な揺らぎの制御や揺らぎの計測は困難であった。本研究では、非天然アミノ酸のタンパク質への部位特異的導入技術を用いて、蛍光共鳴エネルギー移動のドナーおよびアクセプターとなる非天然アミノ酸をタンパク質の特定部位へ導入することで、タンパク質の構造揺らぎを計測するとともに、さらに20種類の天然アミノ酸に加えてそれらの類似体となる様々な非天然アミノ酸や修飾アミノ酸を導入することで、揺らぎを制御しつつ計測できる手法を確立することを目的として計画した。
