Firefly luciferase catalyzes the emission of yellow-green light and the emission color changes to red by the substitution of an amino acid. We determine the Tyr257, Asn231 and Arg220 mutants. Tyr257 mutants decreased the hydrophobic interaction in the active site. Asn231 and Arg220 mutants broak the hydrogen-bonding network of oxyluciferin binding region. In the result, it is supposed that the change of the emission color is caused by the energy loss of the excited oxyluciferin.
