Research Abstract

遺伝情報発現制御の場を支える分子基盤、構造基盤を理解するために、染色体DNA上に形成される機能構造体の構成蛋白質をプロテオミクスにより同定し、その機能を明らかにすることを目的とした。具体的には、セントロメアやテロメアに恒常的に存在するヘテロクロマチン(constitutive heterochromatin)の構成因子であるHP1(heterochromatin protein 1)に結合する因子100種類をヒト細胞から同定しており、それらの因子の機能解析をとおして遺伝情報発現制御の場を支える分子基盤、構造基盤の解明を目指す。特に、ヘテロクロマチンが持つ常に凝縮した染色体構造、発現制御を担う機能構造体との相互作用、染色体の空間的配置、の3者間にあるクロストークを明らかにし、ヘテロクロマチンが関与する染色体の折りたたみや空間配置による遺伝子発現制御の理解を目的としている。
