Research Abstract

生物は様々な脂溶性化合物に曝されており、これらの化合物は健康や寿命に大きく影響しているものと考えられる。こうした化合物には、環境中の様々な有機化合物だけでなく、ポリフェノールやイソチオシアネート化合物などの機能性食品成分、酸化ストレスにおいて生成される酸化脂肪酸なども含まれる。こうした脂溶性化合物は細胞膜あるいは細胞内受容体に作用し、受容体刺激による適応応答の誘導に関与することが明らかとなってきている。例えば、核内受容体の天然リガンドとして様々なアラキドン酸代謝産物が同定されているが、これらのリガンドは脂肪細胞分化やマクロファージ泡沫化抑制などの活性を示す。本研究では、食品成分を含めた内因性および外因性の脂溶性物質に関し、増殖因子などによる受容体シグナリングに与える影響を解析する。特に、脂溶性活性種の中に受容体刺激とともに活性酸素産生を活性化するものを見いだしており、センサータンパク質への直接的相互作用を介したシグナリング機構を解析する。
