Research Abstract

ATPの加水分解エネルギーの物理的実体はいったい何なのか?この究極的問題を解明するため、ATP分解にともなう水の動的状態の変化を調べる。本研究では、「構造」、「ダイナミクス」、「エネルギー論」を統合するエネルギー変換理論の確立を目指して、ハイパーモバイル水(HMW)を軸にした水和の分子論を展開する。具体的には、誘電緩和法によるATPおよびその加水分解産物ADPなどの水和特性を測定し、HMWを含め水和各成分の量的比率を求める。アクチンおよびミオシンの水和状態の特性を誘電緩和法による回転緩和情報とパルス磁場勾配スピンエコー(PFG-SE) NMRによる並進拡散係数から明らかにし、アクトミオシンによるATP加水分解過程の水和特性変化を評価する。また、ATP加水分解過程の反応熱を実験的に再検討する。これらの結果をもとに、溶液化学・統計熱力学の理論家と共同して自由エネルギー変化の内部構成(内部エネルギー項、エントロピー項)を解析し、ATP加水分解エネルギーの物理的描像を構築する。
