Research Abstract

ATPはその加水分解に際して得られる自由エネルギーを通して生体内のエネルギー通貨とも言える役割を果たしている.ATPの加水分解の自由エネルギー変化は水中でおこるため反応前後の水との相互作用の変化の影響を強く受けており,蛋白質がATPを分解して機能する場合も蛋白質と水の相互作用が重要な影響を及ぼす.本研究ではF1-ATPによるATPの加水分解自由エネルギーと回転運動の化学力学変換のメカニズムの理解を出力を定量的に評価するための外部トルク印可システムを用いて,エネルギーの入出力関係を正確に見積るとともに種々のヌクレオチド結合の熱力学的パラメーターを求めて,さらに非水溶媒などを利用してとともに、水中でATPを加水分解して働く分子機械特有のダイナミクスに対する水和の効果を明らかにする.
