Outline of Annual Research Achievements

これまでミクログリアが全身炎症に伴って、血管に遊走し、血液脳関門の透過性に寄与することを明らかにしている(Haruwaka et al., 2019)。さらにミクログリアの体循環系との相関を詳細に検証するために、Toll様受容体7(TLR7)を経皮的に塗布する全身性エリテマトーデスモデルを用いた。本モデルは8週齢マウスから耳にTLR7アゴニストを塗布し、22週齢でdsDNA抗体陽性および脾腫を認める。この期間におけるミクログリアの挙動を、2光子顕微鏡を用いた生体イメージングで毎週同一部位を撮像することによって追跡したところ塗布後10-11週(18-19週齢)においてミクログリアの血管への遊走を認め、血管関連ミクログリアの増加を認めた。これらの分子基盤を検索するため、ミクログリアのRNAシークエンスを行ったところ、Th17関連因子およびCXCL10, CCL5さらにはミクログリアの遊走因子の発現増加を認めた。これに伴ってリンパ球の脳内浸潤の増加を認めた。これらのミクログリアのサブタイプを現在検証中である。さらに神経疾患であるアルツハイマー型認知症における初期のシナプス脱落に対するミクログリアの寄与を領域からの投射ごとに分けて検証し、高次野からの投射を受けるシナプスの選択的脱落にミクログリアが寄与することを明らかにしている。またくも膜下腔におけるマクロファージのアミロイドベータに対する免疫応答を2光子顕微鏡で可視化している。開発したミクログリア特異的に色可変タンパク質が発現するマウスを用いて、生体イメージング情報とトランスクリプトーム情報を統合し病態における変化を明らかにしていく。

Strategy for Future Research Activity

さらに今後研究を進め、ミクログリアの動態とトランスクリプトーム情報を統合し、このリンパ球浸潤への寄与を明らかにしていく予定である。これで論文投稿としていく。さらにアルツハイマー型認知症モデルのミクログリアに対しては開発したミクログリア特異的に色可変タンパク質が発現するマウスを用いて、生体イメージング情報とトランスクリプトーム情報を統合し病態における変化を明らかにしていく
