Outline of Annual Research Achievements

本計画研究では、1分子蛍光イメージング(前島・日比野)、ゲノムHi-CO解析(谷口)、高速AFM・DNA配列結合ポリアミドPIP(杉山)を用いて、ゲノムの構造単位で あるヌクレオソームレベルでのクロマチン構造と動態の観点から、ゲノムモダリティを理解することを目的とする。2022年度、計画代表者前島らは、ヒト生細胞において、発現させたH2B-HaloTagを介してヌクレオソームを非常にまばらに蛍光ラベルし、斜光照明顕微鏡法で単一ヌクレオソームの動態を検出した。そして間期クロマチンの細胞周期におけるヌクレオソームの局所的なゆらぎを観察した。間期ではDNAの量は二倍になり、DNAを収納する核の大きさも二倍以上になる。これまで、このような核内の環境の変化は、クロマチンの動きに大きな影響を与えるとされてきた。しかし本研究によってヌクレオソームのゆらぎの程度は、DNAの量や核の大きさの変化に影響されず、間期でほぼ一定であることを示した(Iida et al. Science Adv.2022)。つぎに代表者前島、分担者日比野らは細胞分裂期の進行に伴うヌクレオソームの局所動態を解析した。これにより、染色体凝縮の進行に伴うクロマチンの物性変化をヌクレオソームの1分子の動きの変化として直接的にとらえた。分担者杉山らはDNAオリガミ法で作成したDNAフレーム中に再構成させたヌクレオソームを配置し、その運動性について高速原子間力顕微鏡で解析した。また配列選択的結合分子であるシリコンローダミンで修飾したピロールイミダゾールポリアミドタンデム4量体を合成し、正細胞において染色体のテロメア領域を近赤外領域で観察した。分担者谷口らは試験管再構成により構築したヌクレオソーム鎖の3次元構造解析の実現を目指し、ヒストン等の必要タンパク質の精製を行うと共に、再構成産物を得るためのプロトコルの構築を進めた。

Strategy for Future Research Activity

計画代表者前島は、ユークロマチンドメインの実体に迫るため、2色の蛍光色素を用いてユークロマチン内の近接した2つのヌクレオソームの動きを同時に観察・比較する。日比野らは1分子・超解像顕微鏡法とタンパク質迅速除去法(AID法)を組み合わせ、コンデンシンIやコンデンシンII,更にはKi67を個別に除去した状態で、ヌクレオソーム動態を1分子計測する。杉山らはピロールーイミダゾールポリアミド(PIP)とDNADNAフレームを用いて遺伝子発現制御について解析を進める。谷口らは試験管再構成により構築したヌクレオソーム鎖の構造解析を行うのに必要な高純度の再構成産物を得るため、生化学条件のさらなる最適化を行うと共に、計算シミュレーションに基づく構造計算法の開発を行う。
