| Research Abstract |
・ﾋｽﾄﾝH2A/H2B 2量体のﾃｲﾙ部分の挙動の多様性がｲｵﾝﾓﾋﾞﾘﾃｨ質量分析（IM-MS）で観測される衝突断面積の分布の多様性を生み出していることが MDｼﾐｭﾚｰｼｮﾝ推測された。そこでﾃｲﾙ部分のｱｾﾁﾙ化およびﾃｲﾙ部分の欠損変異体を調製し構造分布への影響を調べた。その結果、荷電状態に関わらず、天然変性領域が、IM-MSでの構造多様性を生み出していることが示唆された。
・天然変性蛋白質PQBP1、ｽﾌﾟﾗｲｼﾝｸﾞ因子U5-15kD、U5-52KのGYFﾄﾞﾒｲﾝから成るﾍﾃﾛ三量体の立体構造をX線構造解析によって決定した。その結果、U5-15kDとU5-52Kは主に水素結合によって結合していることがわかった。また、表面ﾌﾟﾗｽﾞﾓﾝ共鳴実験により、PQBP1のYxxPxxVLﾓﾁｰﾌとU5-52KのGYFﾄﾞﾒｲﾝが同時にU5-15kDに結合することを示した。
・Sox2 HMGﾄﾞﾒｲﾝは、37℃で第３ﾍﾘｯｸｽを除いてｱﾝﾌｫｰﾙﾄﾞしている。37℃におけるSox2の構造ゆらぎをNMRで解析したところ、第３ﾍﾘｯｸｽも過渡的にｱﾝﾌｫｰﾙﾄﾞしていた。またDNAに対する滴定実験では、非特異的なDNA配列と特異的な配列ともに同じような化学ｼﾌﾄの変化が見られ、さらに非特異的なDNA配列を加えると、過渡的に大きく構造変化していた。以上より、Sox2は、特異的なDNAとの結合構造に似た構造で、かつ揺らぎながらDNA上をｽﾗｲﾄﾞしているものと考えられる。
・組換えﾋｽﾄﾝのH2A/H2B2量体中の構造をNMRとMD計算で解析した。ﾇｸﾚｵｿｰﾑｺｱ中の構造と比較ではH2Bの骨格構造は同じであったがH2AのN末とC末のαﾍﾘｯｸｽが2量体中では壊れていた。ﾋｽﾄﾝｼｬﾍﾟﾛﾝのﾋﾄNAP1のC末の天然変性状態の酸性ﾄﾞﾒｲﾝはH2A/H2Bは特異的に結合し、結合に伴ってH2AのN末のαﾍﾘｯｸｽの形成が誘導された。またﾋﾄNAP1の酸性ﾄﾞﾒｲﾝとの相互作用領域がH2A/H2B上に2箇所存在した。
